Abstract
Although medicinal plants have been historically used for diabetes treatment throughout the world, few of them have been validated by scientific criteria. Diabetes mellitus is a metabolic disorder manifests as elevated blood glucose levels when the body cannot create sufficient amounts of insulin or utilize the insulin produced. This may be due to the free radicals present in the body. The antioxidants help to neutralize the free radicals by donating electron and thus protect from various oxidative stress diseases. The aim of this work is to evaluate the preliminary phytochemical screening, in vitro antioxidant study, in vitro and in vivo anti-diabetic studies of Garcinia pedunculata (GP) leaf extract which have been used as tools to investigate the mechanism of action of drugs with potential antidiabetic property. The antidiabetic potential was evaluated by using Streptozotocin(STZ)-induced diabetic wistar rat model (in vivo) and inhibition of α-amylase and α-glucosidase activity (in vitro). Antioxidant activity was assessed in vitro. Moreover the total phenolic and flavonoid content was also determined. Administration of hydroalcoholic extract of Garcinia pedunculata shows inhibition of α-amylase and α-glucosidase enzymes. There is significant reduction of blood glucose levels in STZ-induced diabetic rats. STZ-induced changes in LDL, HDL, ALT, AST, BID, and Cholesterol levels were significantly restored by treatment with hydroalcoholic extract. Therefore, it has potential as a food supplement to reduce diabetes caused due to oxidative stress.
